氧化应激反应及细胞凋亡与早期压疮形成的关系研究 被引量:15

The role of oxidative stress and apoptosis in the progress of early stage pressure ulcer

摘要 目的探讨早期压疮形成过程中氧化应激反应及细胞凋亡发生的作用机制及其相关性,为临床早期压疮的有效防治寻找实证依据。方法复制前期制作成功的早期压疮动物模型,将36只SD大鼠随机均分为3组,每组12只,均行10%水合氯醛麻醉后,实验A、B两组采用特制压力装置对大鼠股薄肌处进行施压。对照组(C组)不予以压力。两个实验组给予同一压强(22.47kPa),但给予不同压力循环,A组予以3个缺血-再灌注循环(3I/R,I=缺血2.0h,R=再灌注0.5h),B组给予5个I/R。实验终点,肉眼观察受压部位皮肤颜色和形态的变化,采集受压组织中心标本后所有大鼠给予安乐死。通过常规石蜡切片、HE染色、TUNEL荧光标记及酶联免疫吸附测定分别观察皮肤肌肉组织病理学变化、肌组织中细胞凋亡指数(AI)、髓过氧化物酶(MPO)和丙二醛(MDA)含量的变化,并分析各指标之间的相关性。结果 MPO和MDA含量的变化:实验组较对照组显著增加(P<0.01),实验组中的B组含量较A组显著增加(P<0.01)。AI:实验组较对照组显著增高(P<0.01),且随着压力循环的增加AI显著增高(P<0.01)。MDA与MPO呈显著正相关(P<0.01);AI分别与MPO及MDA呈显著正相关(P<0.01)。结论慢性缺血缺氧导致的氧化应激反应及细胞凋亡可能是启动早期压疮发生的分子调控机制之一,并可能是促使其不可逆发展的潜在关键因素。
