摘要 目的:检测糖尿病大鼠内脏脂肪组织中内脏脂肪素(visfatin)mRNA表达及血清visfatin水平,探讨visfatin对糖尿病相关因素的影响。方法:检测糖尿病(DM)大鼠模型内脏脂肪组织visfatinmRNA表达及血清visfatin水平;观察糖尿病大鼠visfatin与空腹血糖(FBG)、空腹胰岛素(Fins)、游离脂肪酸(FFA)、甘油三酯(TG)、总胆固醇(TC)水平、胰岛素抵抗指数(HOMA-IR)及胰岛素敏感指数(ISI)的关系。结果:糖尿病大鼠内脏脂肪组织visfatinmRNA表达及血清visfatin较空白对照大鼠显著增高(P<0.05);糖尿病大鼠血清FFA、TG、Fins、HOMA-IR较空白组大鼠显著增高,ISI显著降低(P<0.05)。血清visfatin与FBG、FFA、HOMA-IR呈显著正相关(P<0.05),与ISI呈显著负相关(P<0.05)。结论:糖尿病大鼠内脏脂肪组织visfatinmRNA表达及血清visfatin水平均明显增高。visfatin的增高可能与糖脂代谢紊乱及胰岛素抵抗状态密切相关。 Objective To detect the expression of visfatin in visceral adipose and serum level of visfatin in rats with diabetic mellitus (DM), and to explore the effects of visfatin on the correlated factors of diabetic mellitus. Methods The expression of visfatin mRNA in visceral adiopose and serum level of visfatin in rats with DM were examined. The relationships between visfatin and fasting blood glucose (FBG), fasting insulin (Fins), free fat acid (FFA), triglyceride (TG), total cholesterol (TC), homeostasis model assessment- insulin resistance index ( HOMA- IR), or insulin sensitivity index (ISI) were observed. Results The expression of visfatin mRNA in visceral adipose and the serum level of visfatin were significantly increased in DM rats than those in normal control rats (P 〈 0.05). The levels of serum FFA,TG,Fins,and HOMA-IR were significantly increased,whilst ISI were decreased significantly in DM rats than those in normal control rats (all P 〈 0.05). Serum level of visfatin was positively correlated with FBG, FFA, and HOMA- IR (P 〈 0.05), and was negatively correlated with ISI (P 〈 0.05). Conclusion The expression of visfatin mRNA in visceral adipose and the serum level of visfatin are significantly increased in DM rats. The increasing of visfatin may be related to the metabolism disorder of glycolipids and insulin resistance
