摘要 目的:探讨临床组织中乳腺癌干细胞含量和球囊形成与临床特性的关系。方法:乳腺癌细胞原代贴壁培养成功后,消化制备单细胞悬液并计数,取恒定密度细胞进行流式检测CD44+/CD24-/low表型乳腺癌干细胞含量,另一部分固定密度细胞行无血清悬浮球囊培养;将肿瘤干细胞含量及成功形成球囊病例数与患者临床特性进行相关性分析。结果:87例乳腺癌病例中,肿瘤干细胞平均含量为0.292±0.158(0.012~0.658),CD44+/CD24-/low细胞含量在ER(-)、ER(+)、PR(-)和PR(+)病例中分别为0.365±0.152、0.242±0.142、0.334±0.163和0.242±0.142;成功形成球囊病例分别占60.0%、32.7%、56.1%和32.6%。经单因素分析显示,ER、PR与肿瘤干细胞含量及能否成功形成球囊明显相关,而HER2、年龄及T分期等与肿瘤干细胞含量及能否成功形成球囊无明显相关。结论:乳腺癌患者癌组织中肿瘤干细胞含量及形成球囊病例数与ER、PR明显相关。 Objectives To investigate the content of human breast cancer stem cells and cases formed mamosphere according to clinical characteristics. Methods Surgically removed breast tumors were dispersed overnight in collagenase Ⅲ, then they were cultured following by flow cytometry and serum-free culture which can enrich cancer stem cells. Finally, the relationship between the prognostic signatures and the content of stem cells and suspension culture were analyzed. Results In the 87 collected patients with breast cancer, average content of cancer stem cells was 0.292 ± 0.158 (0.012 ±0.658), univariate analyses indicated that the conten of CD44+/CD24-ow cells in patients with ER negative or PR negative were significantly higher (ER negative,0.365 ±0.152; ER positive,0.242 ± 0.142;PR negative,0.334 ± 0.163;PR positive,0.242 ± 0.142).The cases which can form mamosphere were more in ER negative or PR negative patients (ER negative,60.0% ; ER positive 32.7% ;PR negative, 56.1% ; PR positive, 32.6% ) (P 〈 0.05 ). However, status of HER2, age and T-stages were not statistically significant factors. Conclusions The content of human breast cancer stem ceils and the cases formed mamosphere are associated with steroid hormone receptor status.
