摘要 目的探讨SSCP法在线粒体DNA分析中的应用及其在法医实际检案中的意义。方法利用PCR方法直接扩增D-LOOP区的HVI16106-16297区域192bp片段作为目的片段,进行SSCP分型。检验150例随机个体、10例正常组织和5例甲醛固定石蜡包埋的组织。结果在150例随机个体中,SSCP检测发现9种单倍型,DP值为0.881。在10例正常组织中,各个组织的SSCP带型完全相同。5例包埋组织的SSCP分型与对照组织一致。结论 SSCP技术分析线粒体DNA有较高的鉴别能力,适用于陈旧降解检材的检验。 Objective To study the application of SSCP technology in the mitochondrial DNA analysis.Methods The highly variable region 16106-16297 in D-loop was amplified by PCR.150 unrelated Liaoning Han individuals,10 normal tissues and 5 formalin-fixed,paraffin-embedded tissues were investigated and analyzed by SSCP.Results In 150 unrelated Liaoning Han individuals,9 haplotypes were found by PCR-SSCP,DP=0.881.The 5 paraffin-embedded tissues displayed alleles different from those of corresponding normal tissue.Conclusion PCR-SSCP technology has a higher ability of mitochondrial DNA identification and is suitable for tests of degraded samples.
