全蝎纯化液对实验性动脉血栓形成血栓素B_2和6酮-前列腺素F_(1α)的影响 被引量:9

Effect of scorpion purified liquid on TXB_2 and 6-Keto-PGF_(1α) during experimental arterial thrombosis in rabbit

摘要 目的观察全蝎纯化液对新西兰白兔颈总动脉血栓形成血栓素B_2(TXB_2)、6酮-前列腺素F1_α(6-Keto-PGF_(1α))的作用。方法采用随机数字表法将50只健康新西兰白兔分成5组:空白组、模型组、全蝎纯化液大剂量组(20 mg/kg)、全蝎纯化液中剂量组(10 mg/kg)和全蝎纯化液小剂量组(5 mg/kg)。耳缘静脉注射全蝎纯化液(空白、模型组注射同等体积的生理盐水)后,采用H_2O_2损伤新西兰白兔颈总动脉制备血栓模型,造模2 h后,采血,称血栓质量、ELISA法检测血浆TXB_2和6-Keto-PGF_(1α)的含量。结果 (1)全蝎纯化液大、中、小剂量组血栓质量比均明显低于模型组[(0.39±0.17)mg/mg、(0.52±0.21)mg/mg、(0.71±0.18)mg/mg比(1.68±0.15)mg/mg,均P<0.01]。(2)与模型组比较,全蝎纯化液大、中、小剂量组均能拮抗TXB_2含量升高[(98.7±23.5)ng/L、(127.6±24.6)n/L、(139.2±31.5)ng/L比(213.5±48.3)ng/L],能抑制6-Keto-PGF_(1α)含量降低[(65.4±21.3)ng/L、(49.8±11.5)ng/L、(41.2±9.6)ng/L比(26.3±6.1)ng/L],差异均有统计学意义(P<0.01)。结论全蝎纯化液能明显降低血浆TXB_2含量、增加血浆6-Keto-PG_(1α)含量,提示全蝎纯化液作用机制可能与维持血浆TXB_2和6-Keto-PGF_(1α)平衡,从而抑制动脉内膜受损后血栓形成有关。 Objective To study the effect of scorpion purified liquid on plasma TXB2 and 6 - Keto - PGF1αduring carotid arterial thrombosis in New Zealand white rabbit. Methods Fifty healthy New Zealand white rabbits were randomly assigned into control group (n = 10), model group (n = 10), high dose (20 mg/kg) (n = 10), medium dose (10 mg/kg) (n = 10) or low dose (5 mg/kg) scorpion purified liquid group. Different dose of scorpion purified liquid or saline were injected via ear vein. The carotid arterial thrombosis rabbit model was prepared by H202 injection. Two hours later, thrombus mass and plasma TXB2 , 6 - Keto - PGF1α were measured. Results ( 1 ) Thrombus mass in high, medium and low dose scorpion purified liquid group were significantly lower than in model group [ (0. 39 ± 0. 17) mg/mg, (0. 52 ± 0. 21 ) mg/mg, (0. 71 ± 0. 18 ) mg/mg vs. ( 1.68 ± 0. 15 ) mg/mg, all P 〈 0. 01] (2) Compared to model group, plasma TXB2 was decreased [ (98.7 ± 23.5 ) ng/L, ( 127.6 ± 24. 6) ng/L, ( 139.2 ± 31.5 ) ng/L vs. (213.5 ± 48.3 ) ng/L ] and6-Keto-PGF1αwasincreased [(65.4±21.3) ng/L, (49.8±11.5) ng/L, (41.2±9.6) ng/Lvs. (26.3±6.1) ng/L, P 〈 0. 01 ] in each dose of scorpion purified liquid group. Conclusions Scorpion purified liquid can prevent thrombus formation through lowering plasma TXB2 level and increasing 6 - Keto - PGF1α level after carotid artery injury in rabbit.
