5-氟尿嘧啶对绒癌JAR细胞凋亡的作用机制

The mechanism of 5-fluorouracil on apoptosis of choriocarcinoma JAR cells

摘要 目的:探讨5-氟尿嘧啶(5-fluorouracil,5-FU)对人类绒毛膜癌细胞株JAR细胞凋亡的作用机制。从分子水平分析绒癌细胞凋亡及凋亡抑制基因XIAP在此凋亡过程中的作用。方法:RT-PCR法检测5-FU对JAR细胞XIAP mRNA表达的影响。结果:5-FU在0~40μg/ml范围内可以呈剂量依赖性下调XIAP基因mRNA的表达,在40~80μg/ml范围内,随着5-FU剂量的增加,下调能力有所减弱,XIAP的表达反而有所增强。结论:5-FU诱导绒癌细胞凋亡可能是其抗肿瘤作用机制之一,5-FU诱导细胞凋亡的机制可能与XIAP mRNA的表达有关。 Objective:To explore the mechanism of 5-fluorouracil on apoptosis of choriocarcinoma JAR cells,analyze the effects of apoptosis of choriocarcinoma cells and inhibitor of apoptosis gene-XIAP in the course of apoptosis of choriocarcinoma JAR cells at the molecular level. Methods:RT-PCR was used to detect the effect of 5-fluorouracil on XIAP mRNA expression in JAR cells. Results:5-fluorouracil within the range of 0~40 μg/ml down-regulated the expression of XIAP mRNA,showing a dose-dependent manner;within the range of 40~80 μg/ml,with the increase of 5-fluorouracil dose,the down-regulation ability decreased,but the expression of XIAP mRNA increased. Conclusion:The apoptosis of choriocarcinoma JAR cells induced by 5-fluorouracil may be one of the anti-tumor mechanisms,the mechanism may be related to the expression of XIAP mRNA
