PBMC参与乙肝自愈免疫反应的蛋白质组学分析

PROTEOMIC ANALYSIS OF PBMC IN SPONTANEOUS CURE OF HEPATITIS B

目的:通过对比分析乙肝血清标志物全阴者与乙肝自限性痊愈者外周血单个核细胞的蛋白质组表达谱,寻找参与乙肝病毒(HBV)免疫反应的关键蛋白质。方法:采用双向电泳技术结合生物信息学分析方法对乙肝血清标志物全阴者及乙肝自限性痊愈者PBMC进行比较蛋白质组学分析。结果:获得了3个有明显规律性变化的差异表达蛋白点,这三个蛋白点共同表现为在乙肝自限性痊愈者中表达量均高于对照组表达量2倍以上。结论:本研究发现的这三个差异表达蛋白质可能直接或间接参与了HBV免疫应答。

Objective：To search the key proteins involved in immune response to HB virus by comparing the protein profiles of peripheral blood mononuclear cell,（PBMC）between HB markers all negative（control）and sole HBs Ab positive（spontaneous cure） healtby volunteers..Methods：Proteins extracted from PBMC were separated by two-dimensional gel electrophoresis（2-DE） and the protein expression profiles were analyzed by comparative proteomics technique to seeh the differential protein spots.Results：Three differential proteins were discovered.All these proteins showed mueh higher expression level in HB spontaneous cure healthy volunteers than in control.Conclusion：The three differential proteins may directly or indirectly involved in the immune response during hepatitis B virus infection.
