曲古抑菌素A刺激肝癌细胞株HepG2的蛋白质组学分析

摘要 目的:通过对比分析曲古抑菌素A刺激肝癌细胞株HepG2 24h与对照组的蛋白质组表达谱,探讨参与曲古抑菌素A抗肿瘤作用的分子机制提供实验依据。方法:采用双向电泳技术结合生物信息学分析方法对曲古抑菌素A刺激肝癌细胞株HepG2 24h后及对照组的细胞全蛋白进行比较蛋白质组学分析。结果:获得了7个有明显规律性变化的差异表达蛋白点,其中一个蛋白点表达量有所升高,6个差异蛋白点表现为明显下调趋势。结论:本研究发现的这些差异表达蛋白质可能直接或间接参与了曲古抑菌素A抗肿瘤的分子机制。 Objective:To explore the anticancer mechanism of trichostatin A(TSA) in HepG2 by comparing the protein profiles of HepG2 with and without stimu lation of Trichostatin A.Methods:Proteins extracted from all HepG2 were separated by two-dimensional gel electrophoresis(2-DE) and the protein expression profiles were analyzed by comparative proteomics technique to seek the differential protein spots.Results:Seven differential proteins were obtained:one protein showed higher expression level(up-regulated)wh:le six other proteins showed lower expression level(down-regulated)in HepG2 aftor stimulated with Trichostatin A.Onclusion:These differential proteins may directly or indirectly involved in the anticancer mechanism of trichostatin A(TSA) in HepG2.
