摘要
为了探讨钙蛋白酶I基因(CAPN1)的多态性,并分析其对优良地方鸡种清远麻鸡和国外引进的隐性白羽鸡肌肉剪切力和系水力等性状的遗传效应,采用PCR-SSCP及测序的方法对CAPN1基因的编码区进行单核苷酸多态性(SNPs)扫描,并使用SAS软件进行最小二乘分析。发现了3个SNPs,分别为外显子5中的T2507C突变和C2546T突变,以及外显子6中的G3535A突变。各基因型与嫩度性状的方差分析结果表明:虽然各基因型在不同品种中表现出的效应存在差异,但趋势一致。结合CAPN1基因的生理功能和已有的研究结果来看,可在特定的群体中将其作为影响鸡肉嫩度性状的候选基因。
To detect the polymorphisms of the coding region of CAPN1 gene as well as its genetic effect on meat tenderness traits in chicken,two chicken breeds,a Chinese indigenous chicken breed Qingyuan partridge chicken and a introduced chicken breed recessive White chicken,were used for this study.Shearing force and water holding capacity of the breast muscle of the two chicken breeds were measured.PCR-SSCP and DNA sequencing were used to identify single nucleotide polymorphisms(SNPs) of the coding region of CAPN1 gene,and the data were analyzed by the least square analysis using SAS program.Three polymorphism sites,T2507C mutation and C2546T mutation in exon 5 and G3535A mutation in exon 6 were found.The least square analysis showed that although the significance levels of the genotypic effects of different loci in different breeds were different,the tendency was coincident.The results indicated that CAPN1 gene could be regarded as a candidate gene affecting meat tenderness traits in special chicken populations.
