摘要
目的通过实验研究观察黄连温胆汤加减对代谢综合征(MS)大鼠内脏脂肪组织中内脏脂肪素(visfatin)mRNA表达及血清visfatin水平;探讨黄连温胆汤加减治疗MS的作用机制。方法采用60只SD大鼠随机分为空白组(n=10)和实验组(n=50)。实验组采用高盐高脂高糖饲料喂养,诱导实验性MS大鼠模型。实验组50只SD大鼠造模成功40只,随机分为模型组(n=10)、黄连温胆汤加减高剂量组(n=10)、黄连温胆汤加减低剂量组加减(n=10)和西药(盐酸二甲双胍)对照组(n=10),空白组、模型组给予同体积生理盐水,分别灌胃给药四周后,RT-PCR检测内脏脂肪组织visfatin mRNA表达,ELISA法测定血清visfatin水平。结果与空白组比较,模型组大鼠内脏脂肪组织visfatin mRNA表达及血清visfatin显著增高(P<0.01);黄连温胆汤加减高剂量组visfatin mRNA表达及血清visfatin水平均明显降低(P<0.05)。结论黄连温胆汤加减能降低大鼠内脏脂肪组织visfatin mRNA的表达及血清visfatin水平,可能是其治疗代谢综合征的机制之一。
Objective To observe the effect of HUANGL1ANWENDAN decoction on expression of visfatin mRNA in fat and visfatin level in blood in order to explore the function of the treatment mechanism of MS.Methods Sixty male SD rats were randomly divided into normal diet group (n=10)and high fat high salt and high glucose diet group (n=50) to induce metabolic syndrome model then metabolic syndrome rats were got 40 successfully, and then these 40 metabolic syndrome rats were randomly divided into model group(n=10), high dose HUANGLIANWENDAN decoction group(n=10), low dose HUANGLIANWENDAN decoction group (n=10) and DMBG group(n=10), normal diet group and model group were feed physiological saline. Expression of visfatin mRNA in fat were inspect by way of RT-PCR and Level of visfatin in blood were observed by way of ELISA after 4weeks. Results In model group expression of visfatin mRNA in fat and level of visfatin in blood are higher than normal diet group(P〈0.01); The data in high dose HUANGLIANWENDAN decoction is depressed evidently (P〈0.05) . Conclusion HUANGLIANWENDAN decoction can reduce the expression of visfatin mRNA in fat and the level of visfatin in. blood which is one of the reason to cure MS.
