摘要
高效、低毒、靶向性的基因载体是基因治疗成功的关键。与病毒载体相比,非病毒载体具有毒性低、免疫原性小、结构改造可实现基因靶向递送、易于生产、可重复应用等优点,其中阳离子聚合物是非病毒载体研究热点之一。本文从基于降低细胞毒性、提高稳定性和转染效率、实现靶向转运等进行的聚合物结构改造策略方面综述了多聚赖氨酸、聚乙烯亚胺、聚甲基丙烯酸酯等阳离子聚合物作为基因载体的研究进展。
A successful gene therapy largely depends on the development of effective,secure and targeting delivery vectors.Compared to the viral vectors,the non-viral vectors have many advantages including lower cytotoxicity,the potential to be administered repeatedly with minimal host immune response,targetability,stability in storage and ease to produce in large quantities.Especially,cationic polymers were given greatly concern.The latest advances on the modification of the polymers to reduce the cytotoxicity,improve the stability and transfection efficiency of the complex,and achieve targeting delivery,are reviewed in this paper.
