不同显色剂对续断皂苷含量测定的影响

摘要 目的考察不同显色剂对紫外-可见分光光度法测定续断总皂苷含量的影响,确定最优显色方法。方法采用紫外-可见分光光度法测定续断总皂苷含量,分别用三种显色法显色,方法A显色剂为浓硫酸:甲醇(4:1);方法B显色剂为5%香草醛-冰乙酸:高氯酸:乙酸乙酯(2:8:10);方法C显色荆为高氯酸。结果方法B专属性较差,方法A和C均适合不同提取物中续断皂苷的9n,4定,方法A加样回收率优于方法C。结论方法A为最佳显色方法。 Objective To investigate the effects of different chromogenic agents on determination of content of total saponins from Dipsacus by UV speetrophotometry, and to establish the optimal chromogenie method. Methods Total saponins were determinated by UV spectrophotometry with three different chromogenic agents, namely sulfuric acid : Methanol (4 : 1 ) ( method A) ; 5 % vanillin - glacial acetic acid : perehloric acid : ethyl acetate (2 : 8 : 10) (method B)and perchloric acid (method C)'. Results Method B performed poorest specificity, and both method A and C were suitable to determine total saponins from different extracts of Dipsacus, but method A was super for higher recovery. Conclusion Method A is the optimal chromogenic method.
