摘要
目的病例报道和文献复习揭示山豆根中毒致人全身肌张力障碍的特殊现象,据此进行动物实验研究,以证实山豆根诱发肌张力障碍模型的假说。方法病例报道和荟萃分析,提出山豆根诱发全身肌张力障碍的神经病学证据和神经影像学证据;进一步动物实验研究,用山豆根诱发SD大鼠出现类似人扭转痉挛的运动障碍症状,并用定量分析的方法对大鼠神经行为学进行观察,神经病理揭示基底节的相应变化。结果中药山豆根中毒可以导致双侧基底节坏死,病人表现为全身性扭转痉挛、姿势异常,语言障碍,吞咽困难。动物实验证实山豆根中毒可以诱发SD大鼠出现类似于人体中毒后的全身肌张力障碍和纹状体细胞的变性坏死。结论中药山豆根中毒可以诱发人和动物全身肌张力障碍,神经行为学和组织病理学证据提示这可能是一种新的肌张力障碍模型。
Objective Special cases with generalized dystonia were reported to reveal that toxicosis of Radix Sophorae Tonkinensis can cause severe generalized dystonia in humans.In view of this clue,to prove the hypothesis,a new model of phenotypic dystonia were conducted on rats.Methods Case cohort in the present study shows the neurological and clinical evidence for the human dystonia caused by Radix Sophorae Tonkinensis.In further experimental study,the movement disorder which looks like human dystonia is induced by Radix Sophorae Tonkinensis in rats.Quantative analysis of neural behaviour and corresponding nruropathological changes in basal ganglia are investigated.Results Poision of Radix Sophorae Tonkinensis can cause the necrosis of basal ganglia of humans,the patients manifestate generalized dytonia,abnormal posture,dysarthria,dysphasia etc.Animal study proves that Radix Sophorae Tonkinensis can induce phenotypic dystonia and bring about the degeneration and death of the nerons in basal ganglia of SD rats.Conclusion The Chinese herb,Radix Sophorae Tonkinensis,can cause generalized dystonia both in humans and rats.The neural behavioral and neuro-pathological evidence sugests a new model of dystonia may be established in this way.
