摘要
目的观察新型肝细胞生长因子肝细胞生成素Cn(HPPCn)在人不同组织和细胞中的表达谱。方法采用Northen印迹和多组织芯片点杂交的方法,在mRNA水平上观察HPPCn在18种人肝源细胞系和非肝源细胞系中的表达水平,以及60种成人和7种胚胎组织中的分布。结果 HPPCn在脑、心、肝和肾中表达水平较高,而在肺、胸腺、横纹肌、结肠等组织中低表达,成年组织中的表达水平高于胚胎组织。HPPCn在肿瘤细胞中表达水平较高,在4种肝癌细胞系中的转录水平是正常肝细胞系L02的2~4倍。结论 HPPCn呈多细胞、多组织分布,在肝癌细胞中的表达水平高于正常肝细胞。
Objective To study the expression profiling of hepatopoetin Cn(HPPCn) in human multiple tissues and cell lines.Methods The expression of HPPCn mRNA was observed in 18 human hepatocyte-derived and non-hepatocyte-derived cell lines,60 adult and 7 fetal tissues by Northen blot or human multiple tissue array.Results The expression of HPPCn mRNA was higher in the brain,heart,liver and kidney,but was lower in the lung,thymus,skeletal muscle and colon in the 60 adult tissues on multiple tissue expression(MTE)array.The level of HPPCn mRNA was higher in adult tissues than in fetal tissues.The expression of HPPCn was apparently high in tumor cells,which was 2 to 4-fold higher in 4 hepatocellular carcinoma(HCC)-derived cell lines than in normal liver cell line L02.Conclusion HPPCn is expressed in various tissues and cells.The expression level of HPPCn is higher in HCC-derived cells than in normal liver cells.
