摘要
目的:观察通冠胶囊对动脉粥样硬化(AS)兔血管内皮功能的影响,探讨通冠胶囊抗动脉粥样硬化的作用和可能机制。方法:健康雄性新西兰白兔36只,按随机数字表法分为空白对照组、高脂对照组、阿托伐他汀组、通冠胶囊大、中、小剂量组,每组6只,空白对照组喂基本饲料,其余各组采用免疫性内皮损伤合并高脂饲料诱导建立AS兔模型,高脂对照组喂高脂饲料,阿托伐他汀组在高脂对照组的基础上加喂阿托伐他汀5mg/kg/d;通冠胶囊小、中、大剂量组在高脂对照组基础上分别加喂通冠胶囊3mg/kg/d、9mg/kg/d及15mg/kg/d,实验周期为6周,各组动物分别于2、6周末行内皮功能超声检测血管内皮舒张功能(FMD)、血管内皮非依赖性舒张功能(NMD)及血脂检测。结果:6周末,通冠胶囊大剂量组、阿托伐他汀组与高脂对照组之间FMD、NMD比较,差异均有显著性意义(P<0.05);通冠胶囊大剂量组NMD、FMD与阿托伐他汀组比较,差异无显著性意义(P>0.05)。结论:通冠胶囊能显著改善动脉粥样硬化兔血管内皮功能,其具有不依赖阿托伐他汀的独立的改善血管内皮功能的作用,而且在改善NMD方面,通冠胶囊优于阿托伐他汀。
Objective. To investigate the therapeutic effect and possible mechanism ot longguan Capsule (IC) on atherosclerosis (AS) through observing the impact of TC on vascular endothelial function of AS rabbits. Methods: Thirty-six healthy male New Zealand white rabbits were randomly divided into blank control group, model group, atorvastatin group, and large-, medium- and small-dose TC groups, six rabbits in each group. The blank control group was fed with the routine forage, and the other groups were given immunity endothelial injury combined with high-fat diet feeding to estalish AS rabbit model. The control group was fed with high fat diet, atorvastatin group was fed with atorvastatin and high fat diet, and TC groups were given high fat diet and small-, medium- and high-dose TC. The course of experiment was six weeks. Flow-mediated dilatation (FMD) and nitroglycerin-mediated dilation (NMD) in each group were tested by ultrasound after two and six weeks. Results: After six weeks, FMD and NMD in high-dose TC group and atorvastatin group were higher than those in the model group, the differences being significant (P 〈 0.05); The differences of NMD and FMD between high-dose TC group and atorvastatin group were not significant (P 〉 0.05). Conclusion: TC can significantly improve endothelial function of atherosclerotic vascular in rabbits, which is independent on the effect of atorvastatin. And TC has better effect than the atorvastatin on improving NMD.
