摘要
目的:为探讨IL2基因导入肝癌浸润淋巴细胞及对其生物学活性的影响。方法:应用逆转录病毒载体介导IL2基因导入人肝癌浸润淋巴细胞(TIL),同时对经转导IL2基因的肝癌浸润淋巴细胞的杀伤活性,增殖能力及表型等进行了观察。结果:转导IL2基因后的肝癌浸润淋巴细胞能表达一定量的IL2,明显高于未转导细胞,但其杀伤肿瘤细胞活性,增殖能力及表型等没有显著变化。结论:应用逆转录病毒载体介导IL2基因导入人肝癌浸润淋巴细胞获得成功,亦有IL2基因产物表达。
Objective: TostudyontransferationwithPZIPnewSVIL2genecomplexsintoTILfromhepatocellularcarcinomaandeffectonbiologicalactivitiesoftransferedIL2geneTIL. Methods: Aneukaryoticexpressionvectorcontaininghumaninterleukin2gene(PZIPnewSVIL2)wastransducedintoTILfromhepatocellularcarcinomabyretroviralvectorgenecomplexsandinsametimephenotype,killingactivityandproliferationoftransducedIL2geneTILwereobserved. Results: TransducedIL2geneTILcouldsecretgenerallevelIL2thatwashigherthanthatofTILsecretion,killingactivity,proliferationandphenotypeoftransducedIL2geneTILwerenosignificantchange. Conclusion: ItwassuccessfulthathumanIL2genewastransducedintoTILandTILcellsoftransducedIL2genebegantosecreteIL2after48 h.
