益气解毒片对鼻咽癌细胞裸鼠植瘤细胞端粒酶抑制作用的实验研究

EXPERIMENTAL STUDY ON THE CONTROL EFFECT OFYIQIJIEDUPIAN ON TELOMERASE ACTIVATY OF IMPLANTED TUMOR CELL IN NASOPHAR YNGEAL CARCINOMA

为了观察益气解毒片在体内对鼻咽癌细胞生长和端粒酶活性的影响。移植鼻咽癌细胞(HNE_1)于BALB/C裸鼠皮下,制备鼻咽癌裸鼠移植瘤模型,肿瘤生长为(1.89±0.37cm^3)。用益气解毒片药液灌胃荷瘤裸鼠30d,测量肿瘤体积和瘤重,同时取瘤组织,用ELISA-PCR法检测瘤组织端粒酶的活性。结果:益气解毒片药液能抑制鼻咽癌裸鼠移植瘤的生长和端粒酶的活性,抑瘤率为82.2±7.56％,用药后瘤组织端粒酶活性受到抑制(A_(450):0.37±0.10),均为阴性。与环磷酰胺对照(抑瘤率:83.25±8.97％,A_(450);0.41±0.09)无明显差异。瘤体积和端粒酶的活性(1.66±0.47cm^3,0.37±0.10),明显小于蒸馏水对照组(1O.21±0.67cm^3,A_(450):1.69±O.21)。益气解毒片抗鼻咽癌作用可能与其抑制鼻咽癌细胞端粒酶的活性有关。

Telomerase activity is present in many kinds of tumor and not detected in normal tissue, and it may serve as a target for therapy.In order to probe the mechanism of antiprohTerative effect for yiqijiedupian to nasopharyngeal carcinoma(NPC) .We made a nude mice model with implanted tumor of NPC(the implanted tumor, 1.89 ± 0.37cm3) .The nude mice were given yiqijiedupian for 30 days and observed the inhibitory effect of the implanted tumor and their telomerase activity by yiqijiedupian. The results showed that the implanted tumor treated by yiqijiedupian was 1.66 ± 0.47cm3 , A450 : 0.37 ± 0.10. The control was 10.21 ± 0.67cm3, A450: 1.69 ± 0.21. The implanted tumors were inhibited by yiqijiedupian (P < 0.05). This showed that the inhibitory effect lanks the down - regulated telomerose activity.
