摘要
以蛹虫草菌丝体发酵液中的菌丝体干重和粗多糖产量为指标,筛选出液体培养基的最适碳源为蔗糖,最适氮源为黄豆粉。通过四因素三水平的正交试验,优化出液体发酵培养基的最佳配方为马铃薯20%,蔗糖5%,黄豆粉2%,磷酸二氢钾0.1%,硫酸镁0.1%,维生素B10.01%。并确定了摇瓶最优发酵工艺条件为装液量100 mL·250mL-1三角瓶,菌龄为72 h,接种量为15%,起始pH 6.0,温度24℃,摇床转速150 r·min-1,最适发酵周期为120 h。
Compared with the dry weight of mycelium and the crude polysaccharide production,the optimal carbon source was sucrose,and the optimal nitrogen source was soybean powder.Through orthogonal experiment with four factors and three levels,the optimal fermentation medium of liquid was potato 20%,sucrose 5%,Soybean meal 2%,KH2PO4 0.1%,MgSO4·7H2O 0.1%,VB1 0.01%.And the best fermentation process control of flask was: filling liquid 100·250 mL-1,age of pure culture72 h,pitching 15%,initial pH 6.0,temperature 24 ℃,stirring rate 150 r·min-1,fermentation 120 h.
