摘要
目的:探讨国内外高通量RNA干扰(RNAi)的技术方法,及其在寻找新的肝癌靶点和治疗方法上的应用。方法:检索Medline及CNKI期刊全文数据库检索系统,以"RNA干扰技术、肿瘤"等为关键词,检索1999-01-2010-12的相关文献。纳入标准:1)高通量筛选;2)药物靶点;3)肝癌。剔除与动物体内实验相关以及技术内容陈旧的文献。根据纳入、剔除标准分析39篇文献。结果:高通量的RNAi技术是一种良好的基因组水平功能性研究工具,可以通过研究肝癌细胞表型来研究基因的内在功能。肝癌病因复杂、易转移,且中国肝癌患者众多,亟待运用高通量的RNAi技术发现针对肝癌新的药物靶点以及更有效的治疗方法。结论:通过文献归纳出,国内外尤其中国在高通量RNA干扰技术在肝癌研究中研究有限,因此运用此技术进行肝癌新的药物靶点和治疗手段的研究具有巨大潜力和重要意义。
OBJECTIVE:To review RNA interference(RANi) technology and the application of high throughput RNAi in identification of novel targets and therapeutic treatments in hepatocellular carcinoma(HCC).METHODS: Medline and CNKI databases were undertaken to identify the relevant articles on high throughput RNAi using in HCC research.Publications from 1999-01 to 2010-12 were searched with the key words of "RNA interference,cancer".Then with the criteria of "high throughput,liver cancer,and drug target",39 articles were collected for summarization in this review.RESULTS: High throughput RNA interference(RNAi),a functional genomics tool,enables us to evaluate the causal relationship between genes and cellular phenotypes.Hepatocellular carcinoma(HCC) is a very aggressive,incurable and highly chemoresistant fatal disease.There are an enormous number of HCC patients in China,making an urgent need to identify new drug targets for HCC as well as more effective therapeutic treatments.CONCLUSIONS: Through the literature study,it concludes that researches on genome-scale RNAi screening in HCC are very limited,especially in China.Therefore,applying this technology has great potentials for advancing pharmacologically relevant targets and agents in HCC research.
