热处理对HeLa细胞增殖、凋亡及HSP70表达的影响 被引量:2

摘要 目的:通过不同温度对HeLa细胞的处理,观察细胞的存活率,细胞凋亡以及HSP70蛋白表达的变化。方法:对HeLa细胞进行培养及不同温度的热处理,采用MTT法测定HeLa细胞的存活,通过免疫组化方法检测HeLa细胞HSP70蛋白表达的变化以及DNA ladder观察HeLa细胞的凋亡变化。结果:45℃高温处理组细胞增殖的抑制最强,HSP70蛋白表达量最高,细胞凋亡程度最高,41℃次之,37℃最低。结论:高温可以增强热休克蛋白HSP70的表达,进而促进细胞的存活,抑制细胞凋亡。 Objetive:To observe the variation of the survival rate of cell,cell apoptosis and variation of HSP70 protein expression through the HeLa cell treated with different temPerature.Method: The HeLa cell culturing and treatment with different temperature,to test the survival of HeLa cell with MTT,to detect the variation of HSP70 protein expression through immunohistochemical method and to observe cell apoptosis through DNA ladder.Results:The strongest suppression of cell proliferation,the highest HSP70 protein expression and the largest cell apoptosis at 45℃ treatment group,then at 41℃,the lowest at 37℃.Conclusion: High temperature may strengthen HSP70 protein expression,then promote cell survival and surpress cell apoptosis.
