摘要
研究了菌龄、酶的配比、酶解时间及渗透压稳定剂等多种因素对玫红穗状霉Spicellum roseum原生质体形成的影响.结果表明,制备玫红穗状霉原生质体的最佳条件为:菌龄30 h,用10 mg/mL纤维素酶+10 mg/mL蜗牛酶+5 mg/mL溶壁酶的混合液酶解,以0.7 mol/L的KC l作渗透压稳定剂,30℃酶解3 h.
The preparation conditions of Spicellum roseum protoplast including incubation time,enzyme system,enzymolysis time and osmotic stabilizer were studied.The optimal preparation conditions of S.roseum protoplast were 30 h mycelium growth,10 mg/mL cellulase,10 mg/mL snailase and 5 mg/mL lywallzyme,3 h reaction at 30 ℃ and 0.7 mol/L KCl as osmotic stabilizer.
