摘要
目的观察艾灸对腹透模型大鼠腹膜结构、血管内皮生长因子(Vascular Endothelial Growth Factor,VEGF)及一氧化氮(Nitrogen Monoxide,NO)的影响。方法将大鼠随机分为正常组、模型组、假透组和艾灸治疗组,采用腹腔注射高糖腹透液(葡萄糖浓度为2.5%)制备大鼠腹透模型。艾灸治疗组以腹腔注射前30 min温和灸神阙穴5 min为治疗方案。应用HE染色观察大鼠腹膜组织结构变化,硝酸还原酶法测定血清NO含量,采用酶联免疫吸附法和Western Blotting测定血清与腹膜组织中VEGF含量。结果 HE染色结果显示模型组大鼠腹膜组织结构欠完整,血管分布不均匀,数量减少,伴管壁明显异常增厚,与模型组比较,艾灸治疗组大鼠腹膜组织结构有明显改善,表现为大鼠腹膜组织完整,血管数量增加,管壁仅见轻度增生。与正常组比较,模型组大鼠血清与腹膜组织中VEGF含量降低,血清中NO含量增加(P<0.05);与模型组比较,艾灸治疗组血清中VEGF含量增加、NO含量降低(均P<0.01),腹膜组织内VEGF表达有增加趋势但无统计学意义。结论艾灸神阙穴能一定程度上降低腹透液引起的腹膜组织结构损伤,增加血清VEGF含量,降低血清NO含量。
Objective To observe the effect of moxibustion on peritoneal morphology,vascular endothelial growth factor(VEGF) and NO in rats after receiving peritoneal dialysis.Methods Rats were randomized into four groups: normal control group,model control group,sham dialysis group and moxibustion group.The peritoneally dialyzed rat models were developed by intraperitoneal injection of high-glucose based dialysate(glucose concentration 2.5%).The moxibustion group received 5-min moxibustion at acupoint Shenque(CV 8) 30 min prior to the intraperitoneal injection.HE staining was used to observe the alterations of rat peritoneal structure,nitric acid reductase method for determining NO content in serum,and enzyme-linked immunosorbent assay(ELISA) and Western blotting were adopted to determine the concentrations of VEGF in serum and peritoneal tissues.Result HE staining showed damaged peritoneal structure,disordered and decreased blood vessels and abnormally thickening blood vessel wall in the model control group.Compared with the model control group,the moxibustion group showed better peritoneal structure,manifested by intact peritoneal tissues,increased blood vessels and slight hyperplasia of blood vessel wall.Compared with the normal control group,notable reduction in the concentration of VEGF in both serum and peritoneal tissue and significant increase in the content of NO in serum were detected in the model control group(P0.05);compared with the model control group,the moxibustion group had significantly higher concentration of VEGF and lower content of NO in serum(both P0.01),while the increase in the concentration of VEGF in peritoneal tissues was not significant.Conclusion Moxibustion at acupoint Shenque(CV 8) attenuates peritoneal dialysis induced damages to peritoneal structure,increases the concentration of VEGF and reduces the content of NO in serum.
