基于遗传算法的卵黄高磷蛋白提取工艺优化 被引量:4

摘要 为了对提取鸡蛋黄中卵黄高磷蛋白的工艺参数进行优化,在单因素的基础上采用二次通用旋转组合设计试验;考察了有机溶剂比例(正己烷/乙醇)和用量对卵黄高磷蛋白提取效果的影响,并基于遗传算法优化出了提取卵黄高磷蛋白的最佳工艺参数;采用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳法(简称SDS-PAGE)测定卵黄高磷蛋白的分子量。结果表明:当有机溶剂比例(正己烷/乙醇)为3.4/1、有机溶剂用量为148mL时,卵黄高磷蛋白的纯度高达81.62%;SDS-PAGE凝胶电泳结果表明:提取出的卵黄高磷蛋白的分子质量在35kD左右。 The quadratic general rotary unitized design was employed to optimize the parameters of extracting phosvitin from egg yolk proceeding after one-factor experimental design. The ratio of n- hexane to ethonal and their total dosages were investigated to evaluate the extraction rate of phosvitin. The optimized technology parameters of extracting phosvitin were obtained based on genetic algorithm. The SDS-polyacrylamide gel electrophoresis (SDS-PAGE) was used to determine the molecular weight (MW) of phosvitin. The results are obtained as follws: ratio of n-hexane to ethonal of 3.4/1 (V/V), dosage of the total organic solvent of 148 mL, the purity of the phosvitin reached 81.62%. The results of SDS-PAGE indicate that the MW of phosvitin was 35 kD.
