白细胞介素-17对人滑膜细胞基质金属蛋白酶-9基因表达的影响

摘要 目的:探讨白细胞介素-17(IL-17)对人滑膜细胞基质金属蛋白酶-9(MMP-9)基因表达的调节作用及意义。方法:收集急性外伤性半月板损伤患者关节滑膜组织(8例),采用组织块培养法进行原代滑膜细胞培养,加入不同浓度的重组人IL-17,培养12,24及36 h后收集细胞并抽提RNA,应用实时PCR检测滑膜细胞MMP-9 mRNA的表达。结果:组织块培养法可以获得高纯度的人滑膜细胞,且表达一定量的MMP-9。以10 ng/m l的IL-17作用滑膜细胞12 h后MMP-9 mRNA表达增加,24 h到达高峰,36 h后明显下降(F=16.06,P<0.05)。以1,10,20 ng/m l的IL-17作用滑膜细胞24 h均上调滑膜细胞MMP-9 mRNA表达,且存在明显的剂量依赖关系,组间差异有统计学意义(F=24.31,P<0.05)。结论:IL-17能够上调人滑膜细胞MMP-9基因的表达,在关节局部炎性机制中可能发挥重要作用。 Objective: To investigate the effect of interleukin-17 on gene expression of matrix metalloproteinase-9(MMP-9) in human fibroblast-like synoviocytes.Methods: Synoviocyte were isolated from synovial tissues of patients with meniscus injury(n=8) and stimulated with recombinant IL-17 for 12,24 h and 36 h.The expression of MMP-9 mRNA in human fibroblast-like synoviocytes was determined by real-time PCR.Results: The isolated cells accorded with the characteristics of fibroblast-like synoviocytes.The MMP-9 mRNA expression in human fibroblast-like synoviocytes which were stimulated by 10 ng/ml IL-17 increased in 12 h,peaked in 24 h,then decreased significantly after 36 h(F=16.06,P0.05).IL-17 could up-regulated the MMP-9 expression in human fibroblast-like synoviocytes stimulated by 1,10 and 20 ng/ml IL-17 for 24 h,and the expression level was dose dependent(F=24.31,P0.05).Conclusion: IL-17 may play an important role in the development of joint inflammation via up-regulating the MMP-9 expression.
