摘要
目的详细说明建立一种操作简便、稳定可靠的局灶性脑缺血再灌注模型的方法与注意事项。方法 24只雄性SD大鼠随机分两组(模型组和假手术组)。参照并改进Koizumi及Zea-Longa腔内线栓法制备大脑中动脉阻断(MCAO)模型。结果模型组大鼠神经缺陷体征明显,梗死灶明显,梗死部位较恒定;假手术组脑组织结构完整,未见缺血性改变,TTC染色无梗死灶。结论用线栓法制备局灶性脑缺血再灌注模型是成功的。
Objective To summarize the experience of establishing a model of focal cerebral ischemia/reperfusion by a simple,stable and reliable method.Methods 24 adult male SD rats were divided into 2 groups in random:sham operated group and ischemia/reperfusion group.The focal cerebral ischemia/reperfusion model in SD rats was established by the modified Koizumi and Zea-Longa method.Results Compared with the sham operated group,the rats in ischemia/reperfusion group developed typical symptoms of neurological impairment and TTC staining showed the pale at the cerebral infarction obviously and the part of it was constant.Conclusion It achieved success to establish a model of focal cerebral ischemia/reperfusion by the suture-occluded method.
