摘要
目的:为缺血疗法在胰腺癌的临床应用提供基础研究资料。方法:在建立人胰腺癌细胞株裸小鼠胰腺原位移植瘤模型的基础上,结扎荷瘤棵鼠胃十二指肠动脉、胰十二指肠下动脉及背胰动脉诱导胰腺右叶区域性缺血,观察缺血对裸小鼠胰腺移植癌群体增殖动力学及细胞周期动力学的影响。结果:缺血组移植癌生长缓慢,倍增时间延长,移植瘤体积、癌细胞增殖指数及蛋白质含量术后3、7、14d均显著低于对照组(P<0.01)。结论:区域性缺血能有效地抑制裸小鼠胰腺移植癌的生长,并抑制癌细胞DNA及蛋白质的合成。
Objective: To provide experimental data for the clinical application of ischemia therapy intreatment of pancreatic cancer. Methods: After the model of transplanted pancreatic cancer was established innude mice with orthotransplantation of human pancreatic qqncer cell line into the pancreas, the ischemia ofthe right lobe of the pancreas was induced with ligation of the gastroduodenal, inferior pancreaticoduodenaland dorsal pancreatic arteries. The effects of the regional ischemia on the populatiDn proliferative and cytocycle kinetics on the pancreatic transplanted cancer were investigated. Results: The.transplanted cancer grewslower and its doubling time Was longer in the ischemic group than in the control. On the 3rd, 7th and l4thday after operation, the size of transplanted cancer, the proliferative index and protein content of the cancercells were significantly lower in the ischemic group than in the control (P<0. 01). ConcIusion: Reghnal ischemia can effectively inhibit the growth of transplanted pancreatic cancer in nude mice and suppress fhe sgnthesis of DNA and protein in the cancer cells.
