以雄鹌鹑为禽类指示动物,探讨全氟辛烷磺酸(PFOS)对禽类的雄性生殖毒性,染毒组饲料中PFOS浓度分别为10和30mg·kg-1。雄鹌鹑从出生后第9天开始连续染毒54d。实验结束时,观察睾丸和附睾脏器系数、精子数、睾丸组织中LDH和SDH酶活性、血清中甲状腺激素T3、T4和促甲状腺分泌激素(TSH)及睾酮(T)浓度,观察睾丸组织病理学改变以及睾丸组织和泄殖腔形态发育状况。结果表明,30mg·kg-1实验组雄鹌鹑体重从染毒第8天至实验结束显著低于对照组(p<0.05),各组均未出现死亡。实验组睾丸和附睾脏器系数及附睾中精子数增均呈下降趋势,睾丸萎缩。睾丸组织中SDH酶活性和血清中T含量随染毒剂量的增加呈下降趋势,30mg·kg-1剂量组与对照组比较有显著差异(p<0.05)。血清中T3和T4含量随染毒剂量的增加呈下降趋势,TSH含量呈升高趋势,其中30mg·kg-1剂量组T4含量与对照组比较具有显著性差异(p<0.05)。染毒组血清及睾丸中PFOS含量随染毒剂量的增加呈递增趋势,其中30mg·kg-1剂量组血清及睾丸中PFOS含量均显著高于10mg·kg-1剂量组(p<0.05)。实验结果表明,PFOS可引起雄鹌鹑睾丸组织结构损伤,导致睾丸组织萎缩,并影响睾丸组织中酶活性及血清中性激素含量,对甲状腺功能产生一定毒性效应。
To approach the reproductive toxicity of perfluorooctane sulfonate(PFOS,C8F17SO3-)to male avians,an oral administration experiment was performed in the male quail that can be applied as animal indicators.The dose of PFOS were 10 and 30 mg kg-1(calculated with PFOS in forage).The quail were administrated with PFOS for 54 days when body weight,the testicular and epididymal viscera coeff icients,the activities of lactate dehydrogenase isoenzymes and sorbitol dehydrogenase,the level of T3,T4,TSH and testosterone(T)in serum,the level of PFOS in serum and testis were evaluated.Morphological changes in testes and spermatogenetic cells were observed in the light microscope.Morphological and histological changes of cloaca were examined.Results indicated that the body weights of male quail in 30 mg·kg-1 dosage group were significantly lower than those in control group after 8 days exposure to PFOS(p0.01),and no mortality was also observed in every group.The testicular and epididymal viscera coefficients and the number of sperm went down and the testis of male quails withered.SDH,serum T levels have gone down and there was a different increase of SDH and T in 30 mg·kg-1 group compared with that of the control.The concentrations of T3 and T4 in serum decreased with the increase of PFOS dose.The level of TSH rose with the increase of PFOS dose,of which T4 in 30 mg·kg-1 increased differently from the control(p0.05).The level of PFOS in serum and testes rose with the increase of PFOS dose and the level of PFOS in 30 mg·kg-1 increased differently from those in 10 mg·kg-1(p0.05).Results of the study suggest that PFOS induce the testis of male quails damaged and induced testis wither,and had adverse effects on the level of activity of enzyme and sex hormone,as well as the function of thyroid.
