重组人红细胞生成素对大鼠脑缺血再灌注损伤后XIAP及Smac蛋白表达的影响 被引量:1

摘要 目的探讨大鼠脑缺血再灌注后线粒体通路第二种天冬氨酸特异性半胱氨酸蛋白酶激活物(Smac)和凋亡抑制蛋白(XIAP)的表达变化及重组人红细胞生成素(rh-EPO)的干预作用机制。方法将大鼠随机分成缺血组、rh-EPO治疗组,大鼠脑缺血再灌注6h、12h、24 h、72h和7 d检测凋亡,免疫组化法检测Smac和XIAP阳性细胞数。结果缺血组神经元凋亡增多,XIAP、Smac蛋白的表达明显上升;rh-EPO能显著降低神经功能症状,减少神经元凋亡,促进XIAP表达,下调Smac表达。结论脑缺血再灌注后脑组织XIAP和Smac蛋白的表达均明显增加,XIAP作为内源性保护蛋白在脑缺血再灌注损伤中发挥重要作用,rh-EPO可能通过促进XIAP并抑制Smac表达保护神经功能。
