复方银思维对散发性老年痴呆模型鼠学习记忆能力及其海马神经元超微结构的影响 被引量:3

摘要 目的探讨中药复方银思维提取物对散发性老年性痴呆(SAD)模型大鼠空间学习记忆障碍的作用及其海马神经元超微形态学基础。方法采用侧脑室注射链脲佐菌素方法制备痴呆模型(ICV-SAD)。实验大鼠分为假手术组、模型组、多奈哌齐对照组及银思维小、中、大剂量组,共6组。于造模后第21天开始治疗,各治疗组给予相应的药物灌胃,模型组及假手术组给予等体积的双蒸水灌胃。治疗结束后,以Morris水迷宫进行大鼠空间学习记忆能力测试,测试完毕取动物海马组织,电镜观察海马神经元超微结构。结果定位航行试验第3天开始,多奈哌齐组,银思维小、中、大剂量组与模型组相比,平均逃避潜伏期及总游泳距离差异有统计学意义(P<0.05或P<0.01)。空间探索试验中,与模型组相比各治疗组第1象限活动时间明显延长,其中以银思维中、大剂量组差异性最为显著(P<0.01)。假手术组大鼠海马神经元轴突内微管结构完好,排列整齐成束状。模型组大鼠海马神经元轴突内微管结构有明显的断裂或溶解现象,多数区域微管消失,排列紊乱。多奈哌齐组与银思维各治疗组大鼠海马神经元轴突内微管排列整齐,无明显断裂溶解,以银思维大、中剂量组效果最好。结论复方银思维能明显改善SAD模型大鼠的空间学习记忆障碍。其作用的超微形态学基础可能是保护海马神经元微管结构,防止轴突损伤。
