摘要
目的分析FTO基因SNP rs9939609多态性与成年女性肥胖及其相关代谢指标的关系。方法随机选取-80℃保存的"2002年中国居民营养与健康状况调查"中1 250名28~60岁汉族成年女性的冻存静脉血,进行全基因组DNA抽提。采用Taqman-MGB探针技术进行FTO SNP rs9939609分型,其中1 245名成功完成分型。利用"2002年中国居民营养与健康状况调查"资料,分析不同基因型与汉族成年女性肥胖等指标之间的关系。结果 FTO SNP rs9939609 A等位基因表达频率为13.6%,AA、AT、TT基因型的表达频率分别为1.77%,23.69%和74.54%,各基因型的频数分布符合Hardy-Weinberg平衡(P=0.80);与TT基因型携带者相比,携带AA/AT基因型的汉族成年女性发生肥胖的危险性显著升高(OR=1.63,95%CI:1.13~2.44),FTO导致肥胖的人群归因危险度百分比(PAR%)为12.1%;携带AA/AT基因型与TT基因型的超重率、身高、体重、体质指数(BMI)、空腹血糖(FBG)、三酰甘油(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、舒张压(SBP)、收缩压(DBP)等指标差异均无统计学意义。结论 FTO基因SNP rs9939609多态性与中国汉族成年女性肥胖存在相关性,携带AA/AT基因型的汉族成年女性发生肥胖的危险性显著高于TT基因型携带者,但未发现对其他代谢指标的影响。
Objective To investigate the role of the FTO SNP rs9939609 polymorphism on obesity and related metabolic traits in Chinese Han adult women. Methods 1 250 Han adult women's blood samples saved in -80 ℃ by 2002 China National Nutrition and Health Survey were randomly selected. Genornic DNA was successfully extracted. Taqman-MGB probe method was used to detect the FTO SNP rs9939609 polymorphism. Among the 1 250 Han adult women, 1 245 women's FTO SNP rs9939609 were successfully genotyped. Results The frequency of rs9939609 allele was 13.5%, and the frequencies of AA, AT, TT genotypes were 1.77%, 23.69%, 74.54%, respectively. The prevalence of obesity among the carriers of AA/AT genotypes was significantly higher than that among those with TT genotype (14.5% vs. 9.4%, P=0.01). Compared to the carrier of TI' genotype, the likelihood of obesity was 1.B3 (95%CI: 1.13-2.44) for the carrier of AA/AT genotype. The population attributable risk of FTO on obesity was 12.1%. There was no significant association between rs9939509 and obesity-related traits including overweight and obesity rate, height, weight, BMI, FBG, TG, TC, HDL-C, LDL-C, SBP and DBP. Conclusion Genetic variation in the FTO gene associates with obesity in Chinese Han adult women, but there is no significant association among rs9939509 and obesity-related traits.
