IgA肾病患者血清聚合IgA1对足细胞增殖的影响 被引量:3

The Effects of aIgA1 from Patients with IgA Nephropathy on Podocyte Proliferation

摘要 目的观察IgA肾病(IgA nephropathy,IgAN)患者与正常人的血清热聚合IgA1(aggregated IgA1,aIgA1)对足细胞增殖的影响。方法收集原发性IgAN患者及健康人的血清,利用层析法分离获得血清单体IgA1(monomeric IgA1,mIgA1),将mIgA1热聚合为aIgA1。利用浓度为0.25、0.5、1.2 mg/ml的患者与正常人的aIgA1分别刺激小鼠MPC5足细胞株,利用MTT法检测aIgA1对足细胞增殖的影响。结果正常组和患者组aIgA1刺激足细胞后细胞MTT吸光度的差异无统计学意义。刺激24 h组与刺激48 h组MTT吸光度的差异也无统计学意义。不同浓度aIgA1刺激组间MTT吸光度的差异有统计学意义P<0.05),相比于不加刺激的阴性对照组(SFM),1、2 mg/ml组的吸光度分别为其0.46和1.66倍。结论足细胞可能会直接对IgA1产生反应,aIgA1可以影响足细胞增殖;低浓度aIgA1抑制足细胞增殖,高浓度aIgA1促进足细胞增殖;患者与正常人的aIgA1对足细胞增殖的影响没有不同。 Objective To explore the effects of aggregated IgA1 (aIgA1) isolated from patients with IgA nephropathy (IgAN) and heahhy control on proliferation of podocyte. Methods Blood samples of patients with primary IgAN and healthy controls were collected.Serum monomeric IgA1 (mIgA1) was isolated with jaealin affinity chromatography and Sephacryl S-200 HR chromatography, then heated to aggregated form (aIgA1). Mouse podocyte (MPC5) was cultured in vitro, then the effects of algA1 on proliferation of podocyte was observed with MTT. Results There wasn't a statistical difference between MTY absorbency of podocyte stimulated by aIgA 1 from patient and healthy control (P=0.352), and there wasn't a statistical difference between 24 hour group and 48 hour group (P=0.556)too. There was a significant difference between the MTr absorbency in different concentration stimulate groups of aIgA1 (P=0.000), compared with SFM, the absorbency of 1 mg/ml group and 2 mg/ml group separately was 0.46 (P=0.033)and 1.66 (P=0.010). Conclusion The results suggest that podocyte could response to IgA1 in direct manner, aIgA1 could affect podocyte proliferation. High concentration of algA1 could promote podoeyte proliferation;The effects on podoeyte proliferation were similar both the algA1 from patient and healthy control.
