摘要
目的:探讨脑源性神经营养因子(BDNF)基因多态性与精神分裂症的关联性。方法:以111例精神分裂症发作期患者与362例正常对照为研究对象,通过TaqM an探针单核苷酸多态性(SNP)基因分型技术对BDNF基因及基因上游10 kb区域的标签SNPs rs6265和rs11030101进行基因分型,比较各组间基因型、等位基因及联合基因型分布频率。结果:精神分裂症患者(n=107)与正常对照(n=357)的SNPs rs6265和rs11030101基因型、等位基因及联合基因型分布频率差异无统计学意义;患者发病年龄、不同临床分型与正常对照的基因型、联合基因型之间比较差异无统计学意义。治疗4周后,SNPrs6265的G/G型、G/A型及A/A型阳性与阴性症状量表(PANSS)阴性分量表分依次为(16.38±8.17)、(12.63±4.32)及(12.60±5.08)(F=3.442,P=0.037)。治疗4周个人和社会功能量表(PSP)评分的升幅依次为(37.60±17.96),(25.25±19.18)及(22.90±15.44)(F=4.129,P=0.020)。结论:精神分裂症患者SNPs rs6265和rs11030101基因多态性与精神分裂症发病年龄、临床分型、急性期症状无关,但rs6265的A/A基因型患者在治疗后阴性症状及社会功能的改善较其他基因型患者显著。
Objective: To investigate the association between brain-derived neurotrophic factor(BDNF) gene polymorphisms and the onset age as well as levels of clinical symptoms in patients with schizophrenia. Method:A hundred and eleven patients were included with informed consent.80 eligible patients entered into the 4-week treatment study and the serum BDNF,the positive and negative syndrome scale(PANSS) and the personal and social performance scale(PSP)were assessed before and after treatment.The BDNF polymorphism(rs6265 and rs11030101)was genotyped using a taqman probe technology.BDNF gene single nucleotide polymorphism(rs6265 and rs11030101)genotypic and allelic distributions were compared with the clinical characters of schizophrenia. Results:The genotypic and allelic distribution of rs6265 and rs11030101 had no relationship with the onset age,gender,type in patients with schizophrenia.The mean scores of PANSS-negative and the improvement scores of the personal and PSP were significantly different among three rs6265 genotypes after 4-week treatment(F=3.442,P=0.037;F=4.129,P=0.020). Conclusion:Schizophrenic patients demonstrate impairments in social function,which could be improved with short-term treatment of antipsychotics.The BDNF gene polymorphism is related to negative symptoms and social function after 4-week antipsychotic treatment.The rs6265 A/A genotype showed significant less negative symptoms and more improvement of social function after treatment.
