雷帕霉素类衍生物依维莫司在恶性血液病中的作用 被引量:1

摘要 针对不同信号通路的分子靶向治疗已成为一种极有发展前景的肿瘤治疗模式。在许多血液系统的恶性疾病中存在mTOR及其相关信号系统异常激活。mTOR抑制剂通过抑制这些信号传导通路而达到抑制细胞增殖、蛋白合成和细胞周期进展的作用。雷帕霉素类衍生物依维莫司(RAD001)是一种新发现的雷帕霉素衍生物,其通过阻断P13K/Akt/mTOR细胞信号传导通路中mTOR的活性而抑制肿瘤细胞增殖、诱导凋亡和逆转细胞耐药,可以作为一种靶向药物用于治疗恶性血液病。本文对依维莫司的发现、作用机制及其在恶性血液病中的作用进行综述。
