泄浊解毒方对溃疡性结肠炎大鼠TLR_4、IκB-α及IL-8表达的影响

Investigating the Effect of Decoction of Xie-zhuo Jie-du on TLR_4,IκB-α and IL-8 in Rats with Ulcerative Colitis

目的:针对溃疡性结肠炎(Ulcerative Colitis,UC)病机组成"泄浊升清,解毒祛瘀"之泄浊解毒方,通过动物实验,观察泄浊解毒方对溃疡性结肠炎大鼠TLR4、IκB-α及IL-8的影响,探讨该方治疗UC可能的作用机制。方法:采用三硝基苯磺酸(2,4,6-trinitrobenzene-sulfonic acid,TNBS)-乙醇联合造模法诱导大鼠溃疡性结肠炎动物模型,实验分为空白组(BG)、模型组(MG)、柳氮磺胺吡啶组(SG)、泄浊解毒方小剂量组(LG)、泄浊解毒方大剂量组(HG)5组。免疫组化技术检测大鼠结肠组织TLR4、IκB-α蛋白水平的表达,放免法检测大鼠血清IL-8的含量。结果:泄浊解毒方可明显降低TLR4、IκB-α蛋白水平的表达及IL-8的含量。结论:泄浊解毒方对溃疡性结肠炎大鼠有良好的治疗作用,其作用机制可能与降低大鼠TLR4、IκB-α的表达及IL-8的含量有关。

Objective： To observe the curative effect of Decoction of Xie-zhuo Jie-du,whose pathogenesis was dispersing turbidity to ascend healthy qi and detoxifying to remove stasis aiming directly at ulcerative colitis,on the expressions of TLR4,IκB-α and IL-8 through animal experiment,and investigate the action mechanism on UC.Method： The animal model of ulcerative colitis was made by trinitro-benzene-sulfonic acid combined with ethanol.All the rats were divided into 5 groups randomly,which were blank group,model group,control group and treatment group of low dose and treatment group of high dose.Then the expressions of TLR4 and IκB-α in colon were detected by immunohistochemistry,and the content of IL-8 in serum was done by radio-immunity.Result： Decoction of Xie-zhuo Jie-du could decrease the expressions of TLR4 and IκB-α markedly.So could the content of IL-8.Conclusion： Decoction of Xie-zhuo Jie-du has good and reliable therapeutical effect on ulcerative colitis,and its mechanism of action may be related to decreasing the expressions of TLR4 and IκB-α and the content of IL-8.
