何首乌中鞣质与二苯乙烯苷不同配比对大鼠肝功能指标的影响 被引量:31

Effects of different ratios of Tannin and Stilbene Glucoside from Polygonum Multiflorum on liver biochemical indexes in rats

摘要 目的研究何首乌提取物鞣质与二苯乙烯苷不同配比对大鼠肝脏生化指标的影响,探讨何首乌肝毒性的可能物质基础。方法 SD大鼠120只,随机分为4组,每组30只。空白对照组(A组)给予1ml/100g的蒸馏水,B组给予鞣质4.4g.ml-1.kg-1,C组给予鞣质(4.4g/kg)与二苯乙烯苷(1.0g/kg)1∶1配比的药物,D组给予鞣质(4.4g/kg)与二苯乙烯苷(1.0g/kg)2∶1配比的药物。各组连续给药90d,恢复15d。分别于给药60d、90d及恢复期15d时颈总动脉取血,生化法检测肝功能相关指标。结果与A组比较,给药60d,各给药组ALT升高,差异具有统计学意义(P<0.01),B组CHE升高及TBIL降低,差异具有统计学意义(P<0.01),B组和C组TP升高,差异具有统计学意义(P<0.01)。与A组比较,给药90d,B组CHE、TP升高和LDH降低,差异具有统计学意义(P<0.01),C组和D组CHE、LDH、ALB降低,差异具有统计学意义(P<0.01)。与A组比较,恢复期15d,B组TP和ALB升高、C组CHE和ALP降低、D组CHE和LDH降低,差异具有统计学意义(P<0.01)。结论长期大剂量灌胃何首乌中提取的鞣质与二苯乙烯苷不同配比、对大鼠肝脏有损害,但鞣质对肝脏的损害在停药后可以恢复。 Objective To research the effects of different ratios of tannin and stilbene glucoside on liver biochemical indexes in rats and to explore the possible material basis of liver toxicity in Polygonum Multiflorum.Methods A total of 120 SD rats were randomly divided into 4 groups: the control group(group A) was given 1ml/100g of distilled water;group B was treated with tannin 4.4g·ml-1·kg-1,group C was given tannin(4.4g/kg) and stilbene glycosides(1.0g/kg) on 1∶1 ratio;group D was given tannin(4.4g/kg) and stilbene glycosides(1.0g/kg) on 2∶1 ratio.The rats were given the appropriate drugs.The rats were treated for 90 days continuously and 15d in recovery.The carotid artery blood samplings were taken at 60d and 90d and 15d in recovery period after administration,to detect the liver function by biochemical indicators.Results Compared with group A,administered for 60d,the ALT in treatment group was increased,the difference was statistically significant(P0.01);the CHE was increased and TBIL was decreased in group B,the difference was statistically significant(P0.01);the TP was increased in group B and group C,the difference was statistically significant(P0.01).Compared with group A,administered for 90d,the CHE and TP were increased but the LDH was decreased in group B,the difference was statistically significant(P0.01).The CHE,LDH and ALB were decreased in group C and group D,the difference was statistically significant(P0.01).Compared with group A,in recovery period,the TP and ALB were increased in group B,the CHE and ALP were decreased in group C;the CHE and LDH were decreased in group D,the difference was statistically significant(P0.01).Conclusion Long-term and large dosage of tannin and stilbene glycosides extracted from Polygonum Multiflorum with different ratios can damage the liver of rats,but the liver damage can be relieved after withdrawal.
