摘要
为评价珊瑚/胶原/重组人工骨形成蛋白-2复合人工骨(简称复合骨)的骨修复能力,将复合骨植入兔颅骨直径15mm的圆形缺损,以单纯珊瑚植入作对照;术后6周、12周取材,通过组织学观察和计算机图像分析,评价其骨修复能力。发现复合骨植入区形成的骨组织量明显多于同期的珊瑚植入区(P〈0.01);术后12周,复合骨完全被成熟的骨组织取代,其骨修复效果明显优于单纯的珊瑚。说明复合骨具有骨传导和骨诱导活性。
By combining coral with collagen and recombinant human bone morphogenetic protein 2(rhBMP 2), coral/collagen/rhBMP 2 composite was obtained. The composite was implanted into rabbit calvarial critical size defects, plain coral used as control. The animals were sacrificed at 6 and 12 weeks respectively after operation, and specimens were removed and examined by histology and image analysis system. Following implantation of the composite, lamellar bone was shown in all coral pores at 6 weeks, and the composite was absorbed and replaced completely by mature bone at 12 weeks. Following implantation of plain coral, lamellar bone was shown in periphery coral pores in the bone defect, while fibrous tissue invased into the central coral pores and bone was not observed there at 6 weeks, and coral was absorbed completely and replaced by bone and fibrous tissue at 12 weeks. The bone repairing ability and effect of the composite were better than those of coral. This study implies that the coral/collagen/rhBMP 2 composite possesses definitively osteoindutive and osteoconductive activity and better bone repairing ability. The composite appears to be an ideal biomaterial for the repair of bone defects.
