摘要
对奉化水蜜桃多酚氧化酶(PPO)的提取分离工艺进行了研究,结果表明,pH值对该酶的提取率影响最大,该酶的最佳提取条件为pH值8.0,提取时间40min,料液比1∶12(m∶V,g∶mL),离子强度0.2mol/L。硫酸铵分级沉淀研究表明,该酶主要在硫酸铵饱和度为50%~80%时发生沉析,故可利用这段浓度范围对该酶进行纯化,其纯化倍数为13.21。同时,水蜜桃PPO对儿茶素表现出很强的亲和性,其酶促反应米氏常数(Km)为0.22mmol/L,最大反应速度(vmax)为251.69U/min。
The extraction of polyphenol oxidase form honeydew peach was studied.According to the results of single factor texts and orthogonal experiment,pH value was the most important factor for the enzyme extraction,and the optimum conditions of extraction were as follows: pH 8.0;the extraction time 40 min;the rate of solid to solution 1∶12;ionic strength 0.2 mol/L.Salting-out of enzyme showed that the enzyme precipitated between 50% and 80% saturation of ammonium sulphate,and the purification fold was 13.21.In addition,PPO showed highly activity to(+)-catechin,and the Km and Vmax values were 0.22 mmol/L and 251.69 U/min,respectively.
