摘要
以慈竹和硬头黄竹等12个竹种为材料,采用RAPD-PCR方法,研究低温胁迫后不同竹种cDNA分子水平上的变化。结果是低温处理后,不同竹种的cDNA经不同引物扩增后,其RAPD-PCR谱带的变化出现4种类型,即有的竹种出现原有RAPD-PCR谱带的缺失、新谱带的出现以及原有谱带强度的增强和减弱,有的竹种RAPD-PCR谱带未发生改变,这表明低温处理后不同竹种在分子水平上确实发生了变化,这种变化与低温胁迫有关,且因竹种而异。
The changes of RAPD-PCR patterns and clustering analysis of cDNA from twelve bamboo species,including Neosinocalamus affinis and Bambusa rigida ect.,under the condition of low temperature were studied.The results showed that the changes of RAPD-PCR patterns of the cDNA differed from species to species.Compared with the control,the variation patterns of deletion or appearance of new of RAPD-PCR bands,strengthening or weakening of RAPD-PCR bands were found.However,no change of RAPD-PCR bands was found in some species.The pattern changes of RAPD-PCR bands were related to the low temperature stress and different bamboo species.
