摘要
目的:研究邻苯二甲酸二乙基己酯(DE-HP)对成年大鼠睾丸前列腺素E2(PGE2)含量及环氧合酶-2(COX-2)mRNA表达的影响。方法:雄性SD成年大鼠40只,随机分成4组,每组10只,设3个染毒剂量组(10、100、500 mg.kg-1.d-1DEHP溶媒为玉米油)和1个对照组(玉米油),饲养4周后处死。用ELISA测定睾丸组织PGE2含量;应用免疫组化法检测40只雄性SD成年大鼠睾丸组织中COX-2的表达及其定位情况;RT-PCR法检测各组睾丸组织COX-2 mRNA表达及其定位情况。结果:实验组睾丸组织中PGE2含量和睾丸组织的COX-2mRNA表达均低于对照组,COX-2则主要表达于精原细胞。结论:DEHP能使成年大鼠睾丸生精细胞功能损伤,使睾丸组织PGE2合成降低,其机制可能与下调COX-2 mRNA表达有关。
AIM : To explore the effects on prostaglandin E-2 and cyclooxygenase-2 mRNA expression of testicular spermatogenic cells in adult rats exposed to di(2-ethylhexyl) phthalate(DEHP).METHODS : 40 male Sprague-Dauley rats were divided into four groups in random: normal control group and 3 groups exposed to DEHP at the dose of 100,200 or 500 mg/kg bodyweight per day by gavage respectively.One month later,the rats were sacrificed.The expression of testicular prostaglandin E-2 was measured by ELISA.The expression level and location of cyclooxygenase-2 and cyclooxygenase-2 mRNA in testicular tissue were detected by immunohistochemistry and RT-PCR,respectively. RESULTS :The expression levels of prostaglandin E-2 and cyclooxygenase-2 of testicular tissue decreased in 3 groups compared with normal control group,cyclooxygenase-2 was mainly expressed in spermatogonia.CONCLUSION : DEHP can damage spermatogenic cells of adult mice,the down-regulating expression of cyclooxygenase-2 mRNA may be involved in it.
