黄连不同部位中5种生物碱的含量比较 被引量:6

Determination of Five Alkaloids in Different Parts of Coptis chinensis Franch. by Ultra Performance Liquid Chromatography

摘要 目的建立超高效液相色谱法同时测定黄连不同部位中5种生物碱含量的方法。方法采用ODS C18柱(50 mm×2.1 mm,1.7μm),乙腈-乙酸铵(0.1 mol/L)水溶液(25∶75,pH为10)为流动相,流速:0.4 m l/m in,检测波长:350 nm。结果在此分离条件下,5种生物碱均分离效果好;线性范围:药根碱0.00070.007μg,r=0.9996;表小檗碱0.00080.008μg,r=0.9995;黄连碱0.00060.006μg,r=0.9992;巴马汀0.0010.01μg,r=0.9998;小檗碱0.002 150.0215μg,r=0.9994。结论该方法操作简便、分析速度快、灵敏度高、精密度和重复性好、结果准确可靠,可用于黄连中5种生物碱的含量测定。 Objective To establish a method for the determination of five alkaloids in different sections of Coptis chinensis Franch.by an ultra performance liquid chromatography method.Methods The ultra performance liquid chromatography was performed on a Beh C18 column(50 mm×2.1 mm,1.7 μm),with a mixture of acetonitrile-water solution of 0.1 mol/L ammonium acetate(25∶75,the pH was adjusted to 10 with aqua ammoniae) as the mobile phase at a flow rate of 0.4 ml/min.The UV detection wavelength was set at 350 nm,and the column temperature was at 25℃.Results Under the above chromatographic conditions,the five alkaloids showed good segregation and a linear relationship.The calibration curves for jatrorrhizine,epiberberine,coptisine,palmatine and berberine were found to be linear within the range of 0.0007-0.007 μg,r=0.9996,0.0008-0.008 μg,r=0.9995,0.0006-0.006 μg,r=0.9992,0.001-0.01 μg,r=0.9998,0.002 15-0.0215 μg,r=0.9994,respectively.Conclusion This method was simple,efficient,highly sensitive,reproducible and reliable.It could be used to evaluate and control the quality of Coptis chinensis Franch.and its preparation.
