摘要
【目的】研究黄芩苷抗心肌肥厚的作用与机制。【方法】昆明小鼠60只,随机分为对照组、模型对照组、黄芩苷高(10 mg/kg)、中(3 mg/kg)、低(1 mg/kg)剂量组。结扎小鼠胸主动脉建立压力负荷心肌肥厚模型,腹腔给予黄芩苷,通过测量全心指数(HW/BW)和左室指数(LVW/BW)评价黄芩苷的抗心肌肥厚作用;分离小鼠心室肌细胞,应用激光共聚焦检测细胞内钙。【结果】模型对照组与空白对照组比较,心脏重量和左心室重量明显增加。与模型对照组比较,高剂量黄芩苷具有明显抑制心脏重量增加和左心室重量增加的作用,其HW/BW和LVW/BW分别为(5.6±0.3)和(3.9±0.2)(P<0.01);中剂量黄芩苷HW/BW和LVW/BW分别为(6.1±0.2)和(4.4±0.3),与模型对照组相比(P<0.05);低剂量黄芩苷有抑制全心重量增加的作用,HW/BW为(6.5±0.2),与模型对照组相比(P<0.05);低剂量黄芩苷具有抑制左心室重量增加的趋势,LVW/BW为(5.0±0.3),但与模型对照组相比(P>0.05),无统计学差异。模型对照组与空白对照组比较,左心室细胞内钙含量较高;高和中剂量黄芩苷组细胞内钙含量较低。【结论】高剂量和中剂量黄芩苷具有抑制心肌肥厚的作用,其机制与细胞内钙含量降低有关。
【Objective】 To study the effect of baicalin mediated antihypertrophic effect and its mechanism.【Methods】A total of 60 Kunming mice were randomly divided into control,model control group,baicalin high(10 mg/kg),medium(3 mg/kg),and low(1 mg/kg) dose group.Thoracic aorta of the mice were ligated to establish pressure load myocardial hypertrophy model.Baicalin was given with different dosage intraperitoneally.Whole heart index(HW/BW) and left ventricular index(LVW/BW) were determined to evaluate the antihypertrophic effect mediated by baicalin.Ventricular cells were isolated and intracellular calcium was detected by confocal laser technique.【Results】Heart weight and left ventricular weight in model control group and control group significantly increased.High-dose baicalin signifieanlly in hibited the increase of weight of heart and left ventricle as compard with model control group.Its HW/BW and LVW/BW was(5.6±0.3) and(3.9±0.2)(P0.01) respectively.Median dose baicalin also inhibited HW/BW(6.1±0.2) and LVW/BW(4.4±0.3) vs model control group increment(P 0.05).Small doses baicalin apparently inhibited HW/BW(6.5±0.2) vs model control group(P 0.05),and in a certain extent suppressed left ventricular weight increases,but was not statistically significant LVW/BW(5.0±0.3) vs model control group(P 0.05).Intracellular calcium content was higher in model control group as compared with blank control group while it was lower in high or median dose baicalin group.【Conclusion】 High and median dose baicalin could inhibit myocardial hypertrophy,its mechanism is relevant to low content of intracellular calcium.
