NF-κB在Caski细胞DEK基因沉默中的作用研究

摘要 目的研究NF-κB在宫颈癌DEK基因沉默中作用。方法实验分转染psiRNA-hHDEK组、转染psiRNA-hH1neo组及未转染组,采用免疫细胞化学染色和western blot方法,观察三组细胞NF-κB的表达情况。结果 psiRNA-hHDEK转染组CaSki细胞NF-κB蛋白表达明显增加;经显微图像分析测定转染psiRNA-hHDEK组阳性细胞百分率明显高于未转染组[(74.2±8.1)%vs(11.7±2.6)%]和转染psiRNA-hH1neo组[(74.2±8.1)%vs(21.6±3.9)%]。结论 DEK基因沉默后CaSki细胞内NF-κB表达上调,DEK负性调控宫颈癌细胞中NF-κB表达,从而发挥其衰老抑制基因及凋亡抑制基因的作用。 Objective To research the effect of NF-κB in the cervical cancer Caski cells by the silence of the DEK gene.Methods The study was divided into the psiRNA-hHDEK transfected group,the psiRNA-hH1neo transfected group and the non transfected group.The NF-κ B level of transfected cells in the three groups was studied by the immunocytochemical staining and Western blot analysis.Results The expression of NF-κ B protein level obviously increased in the group of the CaSKi cells which were transfected psiRNA hHDEK.The percentage of positive cells in the psiRNA hHDEK transfected group obviously increased than that in the untransfected groups and the psiRNA-hH1neo transfected group[(74.2±8.1)%vs(11.7±2.6)%,(74.2±8.1)% vs(21.6±3.9)%].Conclusions The expression of NF-κB is up-regulated after DEK gene silencing in CaSki cells.It means that DEK gene is the negative regulator of NF-κB transcription factor which plays a double regulatory role in cervical cancer by cell senescence and apoptosis.
