摘要 目的:探讨2型糖尿病患者外周血淋巴细胞氧化应激改变对淋巴细胞功能的影响。方法:纳入2型糖尿病患者29例(A组)和健康对照者31例(B组),分离各组样本外周血淋巴细胞,检测淋巴细胞内氧化型谷胱甘肽(GSSH)和还原型谷胱甘肽(GSH);作外周血淋巴细胞涂片,先以活性氧荧光染液处理涂片,观察荧光强度,继以糖原染色(PAS)计算外周血淋巴细胞阳性率;体外以植物血凝素(PHA)刺激分离得到的淋巴细胞并检测其增殖活性。结果:A组淋巴细胞GSSH/GSH为1.69±0.73,B组为1.14±0.63,两组比较有统计学差异(P<0.05)。A组淋巴细胞PAS阳性率为(31±9)%,B组为(25±7)%,两者有统计学差异(P<0.05)。在PHA刺激下淋巴细胞的增殖活性A组为0.61±0.21,B组0.37±0.10,两组比较有统计学差异(P<0.01)。结论:2型糖尿病患者外周血淋巴细胞GSSH/GSH比值增高和PAS阳性率较高与细胞内氧化应激有关;通过检测2型糖尿病患者糖化血红蛋白可间接反映淋巴细胞的转化活性。 Objective To explore the effect of oxidative stress in peripheral lymphocytes on the cell function. Methods Peripheral blood lymphocytes were separated from 29 diabetic patients (group A) and 31 healthy people (group B ). Intracellular GSH and GSSH were detected. Lymphocyte smears were made and then treated with Schiff staining. The positive rate of peripheral lymphocytes was calculated using PAS. The proliferation activity of the lymphocytes cultured in R1640 medium with phytohemagglutin was detected. Results GSSH/GSH was significantly higher in group A than in group B (1.69±0.73 vs. 1.14±0.63, P 〈 0.05). The positive rate of PAS differed significantly between group A and group B [(31± 9)% vs. (25±7)%, P 〈 0.05 ]. There was a significant difference in the proliferation activity of lymphocytes between the two groups (0.61±0.21 vs. 0.37±0.10, P 〈 0.01). Conclusions An increased GSSH/GSH and a higher rate of PAS are associated with intracellular oxidative stress in type 2 diabetic patients. Detection of HbAlc may indirectly reflect the proliferation activity of lymphocytes.
