摘要
[目的]探讨非选择性的醛固酮受体拮抗剂——螺内酯对2型糖尿病大鼠胰岛素抵抗的影响.[方法]高糖高脂喂养加小剂量链脲佐菌素(streptozotocin,STZ)注射的方法建立2型糖尿病大鼠模型,将成模的T2DM大鼠随机分为糖尿病对照组(DM-C组)和螺内酯(SPI)干预组(DM-S组),继续予以高糖高脂饲料喂养,正常对照(NC)组大鼠继续予以普通饲料喂养,其中DM-S组予以SPI 140 mg/(kg·d)灌胃,NC组和DM-C组灌以等量蒸馏水.每两周检测大鼠的体重、血糖水平,第16周末处死大鼠,收集血液标本,检测空腹血糖(FBG)、空腹胰岛素(FINS)、糖化血红蛋白(HbA1c)、甘油三酯(TG)、胆固醇(TC)、血钾及醛固酮(ALD)水平,计算稳态模型评估的胰岛素抵抗指数(HOMA-IR).[结果]DM-C组和DM-S组大鼠的HOMA-IR、FBG、TC、TG均明显高于NC组(均P<0.01),DM-S组TG水平低于DM-C组(P<0.05).DM-C组和DM-S组大鼠的血ALD水平高于NC组(P<0.05),但DM-C组和DM-S组之间无显著性差异(P>0.05).其余指标两组比较差异无统计学意义.三组之间血钾水平无统计学差异(P>0.05).[结论]SPI对2型糖尿病大鼠稳态模型评估的HOMA-IR没有影响,但可以降低TG水平.
[Objective] To investigate the effect of nonselective aldosterone(ALD) antagonists spironolactone on insulin resistance in type 2 diabetic(T2DM) rats. [Methods]The model of T2DM rat was established by feeding with high-sucrose high-fat diet and injecting with low-dose streptozotocin(STZ). T2DM rats were randomly divided into diabetes mellitus control group(group DM-C) and spironolactone group(group DM S), and fed with high-sucrose-high fat diet continuously. The rats in normal control group(group NC) were fed with normal diet. Group DM-S was given 40 mg/(kg · day) spironolactone by lavage. Group NC and group DM-C were lavaged with isovolumic distilled water. Body weight and blood glucose of rats were determined once two weeks. The rats were killed at the end of 16 weeks. Blood samples were collected. Fasting blood glu cose(FBG), fasting insulin(FINS), HbAlc, triglyceride(TG), total cholesterol(TC), blood potassium and ALD were measured. Homeostatic model assessment(HOMA) insulin resistance index(HOMA-IR) was calculated. [Results] HOMA-IR, FBG, TC and TG in group DM-C and group DM-S were significantly higher than those in group NC(all P 〈0.01). TG level in group DM-S was lower than that in group DM-C( P 〈 0.05). There was no statistical significance in other indexes between group DM-C and group DM-S. Blood ALD in group DM-C and group DM S was higher than that in group NC( P 〈0.05), but there was no significant difference between group DM-S and group DM C( P 〉0.05). There was also no statistical significance in blood potassium among three groups( P 〉0.05). [Conclusion] Spironolactone has no influence on HOMA-IR in T2DM rats, but can reduce the TG level.
