摘要
目的 该研究旨在建立大鼠脑出血的动物模型的基础上观察VEGF在出血灶周脑组织中的细胞定位和动态表达.从而探讨VEGF的表达在脑出血后继发性脑组织损伤及修复中的作用.方法 应用脑内立体定向仪,采用二步法自体血注入制备大鼠脑出血模型.Wistar大鼠48只,随机分为脑出血组和对照组,分别于6h、24h、2d、3d、5d、7d后处死,免疫组织化学染色显示脑内VEGF表达变化;显微镜下计数脑内VEGF表达阳性的细胞.结果 脑出血组VEGF表达阳性细胞数随时间逐渐增多,随着出血时间延长,表达VEGF的内皮细胞数量逐渐增多,存在较多微血管和血管样结构.结论 脑出血灶周VEGF的表达增加,7d达高峰,但主要在血管内皮细胞表达,提示VEGF可诱导新生血管的生成,对脑出血后继发性缺血性损伤起修复作用.
Objective To build up the animal model of rats with ICH and to confirm the cell location of VEGF-positive cells in perihematoma and the expression of VEGF. Methods ICH model were induced by two-step.48 rats were randomly divided into two groups,control and experimental ICH groups.The immunohistochemical method and computer image analysis were performed in rats brain by using antibody of VEGE Result There was up-regulation of VEGF in the rat brain tissues after ICH compared with that of the control group.At 7d,there were also many positive cells,mainly endothelial cells,and there were lots of vascular structure. Conclusions The upregulated expression of VEGF induce angiogenesis in perihematoma after ICH in rats,VEGF might play an important role in the recovery of secondary injury brain tissue after ICH.
