摘要 目的 探讨Nemo-like kinase(NLK)在胶质瘤细胞U87MG凋亡中的作用.方法 建立NLK的过表达和小干扰质粒干扰NLK在胶质瘤细胞株U87MG中的表达.CCK-8和细胞免疫印迹的方法检测NLK和胶质瘤细胞U87MG凋亡指标caspase-3的关系.结果 构建的过表达和小干扰质粒能明显地影响NLK的表达.CCK-8和细胞免疫印迹的结果显示NLK能够诱导胶质瘤细胞U87MG的凋亡.结论 NLK能够通过激活经典的凋亡途径而诱导胶质瘤细胞U87MG的凋亡. Objective To investigate function of Nemo-like kinase(NLK) on U87MG cell apoptosis. Methods Over-expression ofNLK plasmid and psilencer 4.1-CMV-NLK small interfering plasmid was established to analyzed the function of NLK in U87MG cell.We analyzed the effect of NLK on cell apoptosis by CCK-8 and Western blot. Results Over-expression of NLK plasmid and psilencer 4.1-CMV-NLK small interfering plasmid could change the expression of NLK in U87MG cell.The results of CCK-8 and Western blot showed that NLK could induce glioma cells apoptosis via activation of caspase-3. Conclusion NLK could induce glioma cells apoptosis via activation of caspases.Gene therapeutic approachesaimed at up-regulate NLK expression could be developed for management of glioma.
