IL-17~+ T细胞及其相关细胞因子在香烟烟熏致小鼠呼吸道炎症中的变化和作用 被引量:7

摘要 目的观察IL-17+T淋巴细胞及相关细胞因子在香烟烟熏导致的呼吸道炎症中的变化,IL-17的细胞来源以及IL-17+T细胞亚型的比例,初步说明其在疾病进程中的作用。方法采用随机数字表法将24只C57/BL6小鼠分为4组:对照组(C),烟熏10 d(CS10)、20 d(CS20)、30 d(CS30)组;HE染色观察肺组织病理变化;ELISA法检测血清和支气管肺泡灌洗液(bronchoalveolar lavage fluid,BLAF)中相应细胞因子IL-17、IL-22和IL-23的表达水平。流式细胞术检测分泌IL-17的Th17细胞和γδT细胞比例。结果随着烟熏时间的延长,小鼠肺组织中淋巴细胞浸润增多、上皮细胞倒伏及支气管壁变厚增生;IL-17、IL-22和IL-23表达量在各烟熏组之间差异有统计学意义(P<0.05);Th17、IL-17+γδT细胞比例较对照组都有不同程度的增高(P<0.05)。结论 IL-17+T细胞及其相关细胞因子可能是烟熏导致的支气管炎症的始发因素之一,其含量的变化对支气管炎的发展和预后都具有重要提示作用。 Objective To observe the change of IL-17+ T cells and related cytokines in the airway inflammation induced by cigarette smoking in mice,and investigate the source of IL-17 and ratios of IL-17 producing cell subtypes in order to explore preliminarily their functions in the progress of the disease.Methods A total of 24 healthy C57/BL6 mice were randomly and equally divided into control group,CS(cigarette smoking),10 days group(1 h in the morning and 1 h in the afternoon for 10 d),CS20 days group and CS30 days group.All mice were sacrificed after last smoking.Pathological changes of their lung was observed by hematoxylin eosin staining.The concentrations of IL-17,IL-23 and IL-22 in their sera and bronchoalveolar lavage fluid(BLAF) were detected by enzyme-linked immunosorbent assay(ELISA).The frequencies of subtype of IL-17+ T cells in the lymphocytes from their lungs were detected by flow cytometry(FCM).Results Compared with control groups,the lung in CS groups showed different pathological changes,including increased lymphocytes infiltration,lodging epithelium,and thickening bronchial wall,and the changes were more serious with the elapse of cigarette smoking period.The concentrations of IL-17,IL-22 and IL-23 in the cigarette smoking groups was higher than those of the control groups(P0.05).The frequencies of IL-17+T cells and IL-17+γδT cells also were also higher than those of the control groups(P0.05).Conclusion IL-17+T and related cytokine may be the beginning factors of bronchitis.Their quantitative changes may indicate bronchitis development and prognosis.
