摘要
为研究生长抑素受体三种亚型(SSTR1-3)在原发性乳腺癌中的分布特性,以[0633α-35S]dATP标记SSTR三种亚型的寡核昔酸为探针,运用原位杂交技术,检测10例乳腺癌组织中SSTR1—3mRNA的表达模式,并用Leica图像分析仪对表达密度作半定量分析。结果表明,SSTR1—3在乳腺癌中的表达模式以表达SSTR2为主(7/10);从SSTR1—3在乳腺癌中的表达密度看,SSTR2高于其它两种亚型(82.3±1.4);与SSTR1、3相比,SSTR2为优势表达亚型(P<0.01).说明SSTR3种亚型在乳腺癌中可同时表达,其分布模式呈异质性,其表达模式存在着较大的个体差异,从表达率及表达密度的半定量分析来看,SSTR2均为主要表达亚型,提示乳腺癌具有接受生长抑素类似物靶向药物治疗的乐观前景。
The objective of this paper is to investigate the characteristics of expression anddistribution of 3 subtypes of somatostatin receptors (SSTRI-3) in breast cancer.With [a-~35S] dATP labeled oligonucleotide of the 3 SSTR subtypes as probe, using insitu hybridization, patterns of mRNA expression were detected in 10 breast cancercases. Additionally, semi-quantitative analysis of the densities of the expression wereestimated by a compreterized image-analysis system. Patterns of SSTR1-3expression in breast cancer were as follows: SSTR2 expression was dominant in 10cases than that of the other 2 subtypes; Therefore, it has positive prospects forsomatostatin analog-oriented agents to be used in treatment of breast cancers.
