摘要
目的本研究通过检测肝素裂合酶在胃癌组织中的表达含量,探讨肝素裂合酶表达与胃癌发展、浸润和转移的关系,为胃癌治疗提供理论依据。方法应用免疫组织化学法(IHC)检测了86例胃癌组织、23例癌旁组织和14例正常组织中肝素裂合酶蛋白的表达。应用流式细胞术(flow cytometry,FCM)的方法定量检测50例胃癌组织、25例癌旁组织和10例切缘正常组织中肝素裂合酶的含量。运用统计软件SPSS 11.0对数据进行统计分析。结果应用IHC法检测86例胃癌组织、23例癌旁组织和14例切缘正常组织中肝素裂合酶阳性表达率分别为79.1%,65.2%和42.9%,三者之间差别有统计学意义(P<0.05)。应用FCM法对50例胃癌组织,25例癌旁组织和10例切缘正常组织标本中的肝素裂合酶的含量进行检测,其荧光指数(fluorescence index,FI)分别为1.86±0.44,1.22±0.45,1.00±0.21。从胃癌→癌旁→正常组织,FI值明显下降,并且三者之间差别有统计学意义(P<0.01)。结论肝素裂合酶在胃癌组织的表达明显高于癌旁组织和正常胃组织,提示肝素裂合酶可能与胃癌的发展有关;肝素裂合酶表达与浸润程度、TNM分期、淋巴结转移相关,提示肝素裂合酶高表达与胃癌的分期、浸润和转移有关。
Objective To investigate the expression of heparanase in gastric carcinoma for its role in carcinogenesis and metastasis of gastric carcinoma and to provide referential indexes to the treatment of gastric carcinoma.Methods The expression of heparanase was examined by SP immunohistochemical stain(IHC) in 86 gastric carcinoma and 23 adjacent carcinoma and 14 normal gastric mucosa.The expression of heparanase in 50 gastric carcinoma and 25 adjacent carcinoma and 10 normal gastric mucosa were quantitatively examined by flow cytometry(FCM).Results Expressions of heparanase in 86 carcinoma and 23 adjacent carcinoma and 14 normal mucosa were detected by IHC,the positive expression rates were 79.1%,65.2% and 42.9%,the expression of heparanase in carcinoma group was higher than that in adjacent carcinoma group and normal mucosa group(P0.01).There were significant difference among 50 gastric carcinoma,25 adjacent carcinoma and 10 normal mucosa quantitatively examined by FCM(P0.01).Fluorescence index(FI) value of heparanase in carcinoma group,adjacent carcinoma group and normal mucosa group was 1.86±0.44,1.22±0.45 and 1.00±0.21,respectively,FI was in descending order as gastric→adjacent→normal tissue,FI value of heparanase in carcinoma group was higher than that in adjacent carcinoma group and normal mucosa group(P0.01).Conclusion The expression of heparanase in gastric carcinoma was significantly higher than that in adjacent and normal mucosa,indicating that heparanase was involved in tumorigenesis progression.There were obvious relations between heparanase and invasion depth,TNM stage and lymph node metastasis,indicating that the high expression of heparanase was significantly related with invasion and metastasis of gastric carcinoma.
