猪水通道蛋白AQP1多克隆抗体制备及其在猪体内的免疫定位 被引量:3

摘要 采用分子克隆技术制备了猪AQP1多克隆抗体,并应用所制备的抗体分析了AQP1在猪体内的表达定位。结果表明:获得了特异性较强、效价较高的猪AQP1多克隆抗体;应用该抗体进行的免疫印迹和免疫组织化学试验显示,AQP1在猪小肠中央乳糜管内皮细胞、小胆管上皮细胞、肾脏的近曲小管上皮细胞以及大脑脉络丛上皮细胞表达。 Rabbit anti-pig AQP1 polyclonal antibody was prepared and immunolocalization of AQP1 in the pig was determined.The results showed: by immuning the rabbit,the polyclonal antibody was successfully prepared.The results of ELISA and western blotting showed that the antibody had high titer(1∶10 000) and specificity.By immunohistochenmical and immunoblotting analyses,porcine AQP1 protein was strongly expressed in the endothelium of central lacteals in villi of small intestine and in the epithelium of small bile ducts,and AQP1 protein was also expressed in the epithelium of kidney proximal tubule and brain chorioid plexus.
